Phase VIII - Quantitative Transcript Confirmation

Microarray technology allows for the detection of transcripts that are differentially regulated in an experiment. Such transcripts (genes) represent candidates for future genetic manipulation; however, microarray technology is not yet adequately quantitative enough, and real time PCR (“RT PCR”) is the method of choice for such verification. ArrayXpress uses RT PCR technology to quantitate the initial amount of the template; most specifically, sensitively and reproducibly. The RT PCR system is based on the detection and quantitation of a fluorescent reporter. This signal increases in direct proportion to the amount of PCR product in a reaction. By recording the amount of fluorescence emission at each cycle, it is possible to monitor the PCR reaction during the exponential phase where the first significant increase in the amount of PCR product correlates to the initial amount of target template, providing a much more quantitative estimation of transcript levels.
